If i upgrade to 5.0.1.

If I do the upgrade to 5.0.1 will I have problems jailbreaking my iPhone? Should I do it or leave it the way it is?

You should upgrade now to ios 5.0.1 because that's the version which can untether jailbreak. Apple is getting ready to release the iOS 5.1 and if you wait and apple release that 5.1 you will not be able to get the 5.0.1 anymore and so no iOS 5 untether jailbreak for you until the dev-team say otherwise. Upgrade your iOS now.
